Kalsiyuma duyarl\u0131 boya fura-2&#8217;nin (Grynkiewicz, 1985) h\u00fccre membran\u0131ndan giri\u015fi, t\u00fcm h\u00fccre s\u00fcspansiyonlar\u0131nda, adherent h\u00fccre pop\u00fclasyonlar\u0131nda, h\u00fccre alt\u0131 d\u00fczeylerindeki [Ca+2]i \u00f6l\u00e7\u00fcm\u00fcnde de\u011fi\u015fikliklere yol a\u00e7t\u0131\u011f\u0131 rapor edilmi\u015ftir (Lambert D., 1992).<\/p>\n<p>Nano boyutlarda, h\u00fccre alt\u0131 d\u00fczeylerdeki alanlardaki Ca+2 sinyali dalgalar\u0131ndan t\u00fcm h\u00fccre Ca+2 sinyali dalgalar\u0131na kadar de\u011fi\u015fen geni\u015f bir skalada, mikro saniyeler i\u00e7erisinde \u00e7ok say\u0131da Ca+2 sinyali farkl\u0131 h\u00fccre tipleri taraf\u0131ndan kullan\u0131l\u0131r (Berridge, 2006; Bootman ve ark., 2001). Bu h\u00fccresel Ca+2 sinyallerinin karakteristikleri, dokuya \u00f6zg\u00fc Ca+2 ta\u015f\u0131ma sistemlerinin ekspresyonuna ba\u011fl\u0131d\u0131r (Berridge ve ark., 2000). H\u00fccresel Ca+2 sinyalizasyon \u00e7e\u015fitlili\u011fi, her t\u00fcrl\u00fc durumda Ca+2 de\u011fi\u015fimlerini izlemek i\u00e7in kullan\u0131labilecek tek bir teknik olmad\u0131\u011f\u0131 anlam\u0131na gelir. Bununla birlikte, floresan ratiometrik Ca+2 sinyali \u00f6l\u00e7\u00fcm teknikleri, h\u00fccresel Ca+2 sinyali yan\u0131tlar\u0131n\u0131 analiz etmek i\u00e7in \u00e7ok y\u00f6nl\u00fc olarak yayg\u0131n bir bi\u00e7imde kullan\u0131lmaktad\u0131r. Tsien ve arkada\u015flar\u0131 taraf\u0131ndan (Tsien ve ark., 1982) ilk defa a\u00e7\u0131klanmas\u0131ndan bu yana, floresan Ca+2 indikat\u00f6rleri farkl\u0131 deneysel ortamlarda bir\u00e7ok \u00e7e\u015fit h\u00fccre modellerinde Ca+2 sinyallerinin ara\u015ft\u0131r\u0131lmas\u0131nda kullan\u0131lmaktad\u0131r. Uygun florometrik \u00f6l\u00e7\u00fcm cihazlar\u0131 ve uygun indikat\u00f6rler kullanarak, canl\u0131 h\u00fccreler \u00fczerinde h\u00fccre alt\u0131 d\u00fczeyler de dahil, y\u00fcksek h\u0131zda, zaman a\u015f\u0131m\u0131na u\u011framadan Ca+2 sinyallerini izlemek m\u00fcmk\u00fcnd\u00fcr.
